Researchers may have unlocked a new methodology to eradicate tumour cells in the brain.
The team of researchers have created Cancer-killing viruses which could transport Stem Cells through the carotid artery, and spread them over to the metastatic Tumours in the brains of clinically pertinent mouse models. They reported that the result of the removal of metastatic Skin Cancer cells from the brains of these preclinical models is an extended survival. The study also defines a technique of linking this therapy with immune checkpoint inhibitors.
As the one who led the study, Khalid Shah, director of the Center for Stem Cell Therapeutics and Imaging (CSTI) in the BWH Department of Neurosurgery, declared that, metastatic Brain Tumours usually from Skin, Breast or Lung Cancers — are the most typically observed Tumours inside the brain and account for almost 40% of advanced melanoma metastases. He added that, “Current therapeutic options for such patients are limited, particularly when there are many metastases.”
The team first developed diverse BRAF (human gene that encodes a protein called B-Raf) wild-type and mutant mouse models which more thoroughly mirrored whatever can be seen in patients—all for their pursuit of innovative and Tumour-focused therapies which could target several metastases in the brain without injuring the nearby tissues.
In order to formulate a possible new treatment, the experts brought about a population of bone marrow-derived mesenchymal Stem Cells filled with oncolytic herpes simplex virus (oHSV), specifically killing the dividing Tumour cells while also saving the normal cells.
An earlier study by Shah and his associates had revealed that dissimilar kinds of Stem Cells were inherently attracted to Tumours in the brain. After first confirming that the injected Stem Cells to the brain will navigate to many metastatic sites and not to those areas without Tumour in their model, the team then injected the oHSV-laden Stem Cells into the carotid arteries of metastasis-bearing mice. The result is a substantially slower progress in Tumour, not to mention a better chance at survival, as opposed to the models which received unchanged Stem Cells.
Shah, who also serves as a professor at Harvard Medical School and a chief faculty member at the Harvard Stem Cell Institute, said that they are presently generating parallel animal models of brain metastasis from other types of Cancer and also the new oncolytic viruses which possess the capacity to exactly kill a wide range of resistant Tumour cells. He concluded that, “We are hopeful that our findings will overcome problems associated with current clinical procedures. This work will have direct implications for designing clinical trials using oncolytic viruses for metastatic Tumours in the brain.”
